Analysis of a new series of SPTs in this study revealed their effects on the DNA cleavage activity of Mycobacterium tuberculosis gyrase. H3D-005722 and its affiliated SPTs showed substantial gyrase inhibition, concomitantly increasing the degree of enzyme-catalyzed double-stranded DNA fracture. The efficacy of these compounds resembled that of fluoroquinolones, including moxifloxacin and ciprofloxacin, while exceeding the efficacy of zoliflodacin, the most advanced SPT in clinical use. All the SPTs exhibited resilience against the most frequent gyrase mutations associated with fluoroquinolone resistance, displaying, in most instances, improved performance against mutant enzymes compared to the wild-type gyrase. Ultimately, the compounds demonstrated a low degree of activity against human topoisomerase II. The data obtained signify the potential of novel SPT analogs to function as antitubercular agents.
Among general anesthetics, sevoflurane (Sevo) is a highly prevalent choice for use in infants and young children. Inhibiting GABAAR and NKCC1 activity shielded the brain from the neurotoxic effects and cognitive impairment caused by sevoflurane. Specifically, bicuculline and bumetanide effectively protect against the sevoflurane-mediated harm to neurons, the compromised formation of myelin, and the resulting cognitive deficiencies in neonatal mice. For the leading cause of global death and disability, ischemic stroke, the necessity for safe and highly potent therapies persists. For ischemic stroke treatment, a transformable, triple-targeting, and ROS-responsive dl-3-n-butylphthalide (NBP) nanotherapy was engineered. A ROS-responsive nanovehicle (OCN) was initially designed using a cyclodextrin-derived component. The result was a pronounced increase in cellular uptake by brain endothelial cells, stemming from a marked decrease in particle size, a transformation of morphology, and a change in surface chemistry induced by the presence of pathological cues. A ROS-responsive and reconfigurable nanoplatform, OCN, exhibited substantially greater brain accumulation compared to a non-responsive nanovehicle in a mouse model of ischemic stroke, thereby amplifying the therapeutic efficacy of the nanotherapy derived from NBP-containing OCN. In OCN molecules equipped with a stroke-homing peptide (SHp), we found a marked rise in transferrin receptor-mediated endocytosis, in addition to their existing ability to target activated neurons. Ischemic stroke in mice exhibited improved distribution of the engineered transformable and triple-targeting SHp-decorated OCN (SON) nanoplatform within the injured brain, significantly localizing within endothelial cells and neurons. The meticulously developed ROS-responsive, transformable, and triple-targeting nanotherapy, bearing the designation (NBP-loaded SON), exhibited impressive neuroprotective results in mice, surpassing the efficacy of the SHp-deficient nanotherapy at a five times higher dose. The nanotherapy, characterized by its bioresponsiveness, transformability, and triple targeting, reduced ischemia/reperfusion-induced endothelial leakiness. This subsequently improved dendritic remodeling and synaptic plasticity in neurons of the damaged brain tissue, leading to better functional recovery. Efficient NBP delivery to the affected brain tissue, targeting damaged endothelium and activated neurons/microglia, and normalization of the pathological microenvironment were crucial to this success. Additionally, early research suggested that the ROS-responsive NBP nanotherapy demonstrated a positive safety record. Therefore, the triple-targeting NBP nanotherapy, demonstrating desirable targeting efficacy, spatiotemporal drug release control, and considerable translational potential, holds substantial promise for precise treatments of ischemic stroke and other brain disorders.

Bamboo-like carbon nanotubes are engineered to integrate both Ni nanoclusters and atomically dispersed Ni-N-C sites (NiNCNT) to catalyze the exclusive conversion of CO2 to CO at consistent, industrially applicable current densities. NiNCNT's performance is enhanced through hydrophobic modulation of gas-liquid-catalyst interphases, resulting in a Faradaic efficiency (FE) for CO generation of up to 993% at a current density of -300 mAcm⁻² (-0.35 V vs reversible hydrogen electrode (RHE)). Our study aimed to assess the ability of polydatin to inhibit stress-induced symptoms of depression and anxiety in a murine model. Mice were classified into groups, encompassing a control group, a chronic unpredictable mild stress (CUMS) exposure group, and a CUMS-treated group with polydatin. Behavioral assays were conducted on mice, which had previously been exposed to CUMS and then treated with polydatin, to determine the presence of depressive-like and anxiety-like behaviors. In the hippocampus and cultured hippocampal neurons, synaptic function was governed by the quantities of brain-derived neurotrophic factor (BDNF), postsynaptic density protein 95 (PSD95), and synaptophysin (SYN). The assessment of dendritic number and length was conducted on cultured hippocampal neurons. To ascertain the effect of polydatin on CUMS-induced hippocampal inflammation and oxidative stress, we measured inflammatory cytokine levels, oxidative stress markers including reactive oxygen species, glutathione peroxidase, catalase, and superoxide dismutase, as well as elements of the Nrf2 signaling pathway. Following polydatin administration, the depressive-like behaviors stemming from CUMS were reduced in forced swimming, tail suspension, and sucrose preference tests, and further reduced anxiety-like behaviors seen in the marble-burying and elevated plus maze tests. The dendrites of hippocampal neurons, cultured from mice undergoing chronic unpredictable mild stress (CUMS), saw an increase in both number and length after polydatin treatment. This treatment also reversed CUMS-induced synaptic deficits by reinstating appropriate levels of BDNF, PSD95, and SYN proteins, as verified in both in vivo and in vitro experiments. Importantly, hippocampal inflammation and oxidative stress stemming from CUMS were counteracted by polydatin, along with the subsequent deactivation of NF-κB and Nrf2 pathways. The presented study indicates polydatin as a potential remedy for affective disorders, its action originating from a reduction in neuroinflammation and oxidative stress. Our current observations regarding polydatin's clinical applications necessitate a deeper examination through further study.
The detrimental effects of atherosclerosis, a common cardiovascular disease, lead to a distressing escalation in morbidity and mortality rates. The pathogenesis of atherosclerosis is profoundly influenced by endothelial dysfunction, which is, in turn, exacerbated by the severe oxidative stress consequences of reactive oxygen species (ROS). The in vitro and in vivo studies provided definitive evidence that Gd/CeO2 nanozymes efficiently scavenged harmful reactive oxygen species at the cellular and histological levels. Gd/CeO2 nanozymes were found to contribute to a considerable reduction in vascular lesions through the reduction of lipid accumulation in macrophages and the suppression of inflammatory factors, consequently inhibiting the progression of atherosclerosis. Gd/CeO2 possesses the capability to act as T1-weighted MRI contrast agents, allowing for the adequate visualization of plaque positions within a living subject. Through these actions, Gd/CeO2 nanostructures might serve as a potential diagnostic and therapeutic nanomedicine for atherosclerosis, specifically induced by reactive oxygen species.
